Overview:

Endocrine therapy (ET) is standard-of care for most estrogen receptor-positive (ER+) metastatic breast cancers (MBC), but nearly all patients will develop therapeutic resistance. Mechanisms of resistance include activation of alternative signaling pathways, which may be addressed by combinatorial strategies, as well as mutation of the estrogen receptor gene, ESR1. Next generation oral selective estrogen receptor degraders (SERDs) have demonstrated activity in ESR1-mutated ER+/HER2- MBC, and have begun to enter the clinic based on pivotal phase 3 data. Other novel endocrine agents in development include new selective estrogen receptor modulators (SERMs), complete estrogen receptor antagonists (CERANs), ER proteolysis targeting chimeras (PROTACs), oral covalent ERα antagonists (SERCAs), and selective androgen receptor modulators (SARMs).

This Medical Crossfire® satellite symposium will be held ancillary to the 2024 annual breast cancer meeting held in San Antonio, Texas, and will provide a forum for recognized experts in the treatment of breast cancer to discuss the latest strategies for managing ER+/HER2- MBC, including new and emerging agents targeting the ER. A mix of didactic presentations with case-based panel discussions will allow in-depth review of real-world patient scenarios integrated with relevant clinical evidence. Discussion topics will include biomarker testing and the dynamics of ESR1 mutation, individualized treatment planning and the evidence-based integration of next-generation oral SERDs, emerging data with investigational ER-targeted agents, and best practices for the management of adverse events.
